How semaglutide is made, and why the manufacturing process matters

Semaglutide is a GLP-1 receptor agonist: a lab-engineered peptide that closely mimics GLP-1, a hormone your gut releases after eating.
The molecule is produced using recombinant DNA technology — bacteria or yeast are given genetic instructions to build the peptide chain, which is then chemically modified to extend its half-life to around one week.
A fatty acid chain is attached to the peptide so it binds to albumin in the bloodstream, slowing breakdown and enabling once-weekly (injection) or once-daily (tablet) dosing.

The science of semaglutide's construction, from peptide chain to finished medicine

Starting point: a hormone your body already makes

The story of semaglutide begins with GLP-1 (glucagon-like peptide-1) a 30-amino-acid hormone secreted by cells in your small intestine when you eat. GLP-1 signals to the brain that food has arrived, slows the stomach emptying its contents, and nudges the pancreas to release insulin in proportion to blood glucose. Useful signals. The problem is that the body degrades native GLP-1 within minutes; it is far too short-lived to be a medicine on its own.

Novo Nordisk's chemists solved this by building a peptide that is structurally very similar to GLP-1 but is not identical to it. One amino acid is substituted (alanine at position 8 replaced with a bulkier group) to block the enzyme that would otherwise chop the molecule apart quickly. That single change is enough to shift the half-life from minutes to days. A common misconception is that semaglutide is somehow extracted from a natural animal source, it is not. The peptide backbone is biosynthetic, assembled to order, and then modified in the laboratory. No animal glands involved.

How the peptide is actually built: recombinant technology and chemical finishing

The peptide chain itself is produced using recombinant DNA technology, the same approach used to manufacture insulin, certain vaccines, and a wide range of modern medicines. Yeast cells (typically Saccharomyces cerevisiae for semaglutide production) are given a DNA sequence that codes for the GLP-1 analogue. The cells read those instructions and assemble the amino acid chain as they grow. The raw peptide is then harvested, purified through multiple chromatography steps, and chemically modified.

That chemical modification is the clever part. A long fatty acid chain (a C18 fatty diacid) is attached to the peptide via a short chemical linker. This fatty acid tail is the reason semaglutide binds reversibly to albumin, the most abundant protein in blood plasma. Albumin acts as a slow-release reservoir: the drug is sheltered from degradation, released gradually, and cleared slowly enough that weekly dosing becomes practical. The entire manufacturing process (fermentation, purification, modification, formulation) takes place in pharmaceutical-grade facilities under strict quality controls, well before the medicine reaches the licensed UK supply chain. Wegovy is manufactured by Novo Nordisk, which operates some of the largest biologics production sites in the world.

From injection to tablet: two very different formulation challenges

Once the active ingredient exists, it still has to reach your tissues in a usable form. For the Wegovy injection pen, the solution is relatively straightforward: semaglutide is dissolved in a buffered liquid and injected subcutaneously once a week. The fatty acid modification does the rest, sustaining therapeutic levels across the week.

The Wegovy tablet, approved by the MHRA on 11 June 2026 as the first oral GLP-1 medicine licensed in the UK for weight management, presented a harder engineering problem. Peptides are ordinarily destroyed in the stomach, acids and enzymes are designed for exactly that. Novo Nordisk's answer was to co-formulate semaglutide with sodium N-(8-[2-hydroxybenzoyl]amino)caprylate, known as SNAC. SNAC is an absorption enhancer that temporarily raises the local pH around the tablet as it dissolves, protecting the peptide from acid degradation and helping it pass through the stomach lining into the bloodstream. The tablet must be taken on an empty stomach with a small amount of plain water and nothing else for at least 30 minutes afterwards, because food and drink interfere with the SNAC mechanism.

The oral route does reduce bioavailability compared with injection, which is why the maintenance dose for the tablet (25mg once daily) is much higher in milligrams than the 2.4mg weekly injection, even though both deliver semaglutide to the same receptors. Why this all matters for anyone thinking about treatment

Understanding what semaglutide is made from is not just academic. The complexity of the manufacturing process is precisely why genuine semaglutide, produced in licensed facilities, inspected by regulators, and distributed through the authorised UK supply chain, cannot credibly be replicated and sold cheaply through social media or unregistered websites. The MHRA has warned repeatedly about counterfeit and unlicensed weight-loss pens circulating online; some have been found to contain insulin rather than the stated ingredient, with serious consequences for people who injected them.
