Starting Mounjaro: what the evidence says about tolerability at the first dose

SURMOUNT-1 (the pivotal trial published in the New England Journal of Medicine, involving 2,539 adults with obesity) found that tirzepatide produced around 20–21% average body-weight reduction at the 15 mg dose over 72 weeks. Injection-related discomfort wasn't a primary endpoint, but dropout attributable to injection-site issues was low across arms. The dose escalation schedule (starting at 2.5 mg for the first four weeks to allow the body to adjust) means most people's first several injections happen while the dose is at its gentlest, giving time to build confidence with the pen before moving up.
